The robot is a representative of a real customer who wants to use your services and you probably wont know it's a robot, just a normal, polite call. It might even save you time since the robots can place a single call and update your information online, so everyone can just check the internet instead of thousands of people calling you directly. Does it matter that it's a robot? Would you rather know it's a robot or think it's a person?

I wouldn't really care, but I think I'd find it awkward -- at least while the technology is new. I'd probably make an added effort to enunciate clearly and avoid complicated words/phrases/expressions. It would be tempting to fuck around with it a bit, too (if I could be 100% sure it's a robot).

You're at work, I doubt you would be paying enough attention to know it's a robot unless it tells you and I'm sure Google would welcome your attempts to trick it, but your manager probably doesn't want you fucking with the customers.
