A burden is a load that is difficult to bear.

I know, but they use it like, "God has given me a burden for drug users," meaning that they feel they have a special call to preach to drug users. Why "burden" or all words?
Persecution complex, and it's always about them even when it isn't (random drug user has zero bearing on their lives personally). Also, it's missions forward to have a "calling" - it's kind of like coming out as gay, they have to do it at some point or risk being seen as just a regular pew warmer.

I've never heard anyone say that.
But if they did I imagine it would be a reference to Matthew 16:24-26
24 Then said Jesus unto his disciples, If any man will come after me, let him deny himself, and take up his cross, and follow me.

It's possible it's a hipster church thing, that doesn't sound normal protestant, you're right.
